Abstract

Introduction

The World Health Organization (WHO) has designated overweight or obesity as an indication of increased risk for heart disease and diabetes, particularly in adolescents, as well as an increased risk for developing cardiovascular disease (CVD), type 2 diabetes, and type 2 diabetes mellitus. Currently, the World Obesity Standards (WOS) for adolescents (which include body mass index (BMI) ≥35 kg/m2 and/or BMI > 30 kg/m2) are in place to ensure adequate weight loss in adults with weight-related health problems and to ensure adequate weight loss in children and adolescents [

].

In the United States, weight gain is considered to be a risk factor for the development of CVD and related to the development of diabetes in adults [

Currently, there are no controlled data on the safety of orlistat in adolescents with a body mass index ≥35 kg/m2 or BMI > 30 kg/m2. The present study was designed to assess the efficacy and safety of orlistat for the treatment of overweight or obese adolescents with a BMI ≥35 kg/m2 or BMI > 30 kg/m2.

Methods

A prospective, open-label, randomized, multicenter, fixed-dose, fixed-treatment study was conducted between March 2007 and October 2008 in the paediatric obesity clinic, Department of Pediatrics, UCL, University of California San Francisco, San Francisco, California, USA. The study was conducted in accordance with the Declaration of Helsinki and the Good Clinical Practice guidelines.

Study design

The study was conducted as a single-centre, open-label, randomized, single-dummy, multi-centre, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, fixed-dose, and fixed-dose, fixed-dose, fixed-dose, in-vitro, in-vivo, in-vivo, in-vivo, in-vivo, and in-vivo treatment of overweight or obese adolescents with a BMI >35 kg/m2 or BMI > 30 kg/m2.

Study protocol was developed and approved by the ethics committee of UCL, and written informed consent was obtained from all patients. All patients were randomized to receive either 120 mg/d Orlistat (30 mg/d) or placebo.
